What is HPLC?
HPLC stands for High-Performance Liquid Chromatography. It is a laboratory analytical technique used to separate, identify, and quantify each component within a mixture. In the context of research peptides, HPLC is used to determine two things: the identity of the compound (is it actually the peptide it claims to be?) and its purity (what percentage of the sample is the target compound, versus impurities or degradation products?).
Why does HPLC testing matter for research peptides?
Research peptides are complex molecules that can degrade or become contaminated during synthesis, storage, or transport. Without independent verification, there is no way to confirm that a vial actually contains what the label claims, or at what purity. HPLC testing provides an objective, reproducible method for verifying both.
A peptide sold without HPLC verification, or from a supplier unwilling to provide test data, should be treated with caution by any researcher.
How to read an HPLC purity result
HPLC results are typically expressed as a percentage — for example, “99.3% purity” means that 99.3% of the sample detected by the analysis corresponds to the target peptide, with the remaining percentage comprised of trace impurities, related substances, or synthesis byproducts.
A Certificate of Analysis (COA) will usually show this figure alongside the batch number, test date, and the specific method used, allowing researchers to trace results back to the exact batch they have received.
How to review an HPLC-tested result
An HPLC-tested label should lead to the underlying method and chromatogram, not replace them. Begin by confirming that the document identifies the same material and batch as the received sample. Review the sample name, analytical date, detector information, column and mobile-phase details where supplied, and the way the purity percentage was calculated.
The principal peak should be interpreted together with the remaining peaks, baseline, integration and method conditions. A large principal peak can support a chromatographic purity statement under that method, but it does not independently establish molecular identity, biological activity or suitability for a particular experiment.
HPLC-tested purity versus identity
HPLC separates detected components according to their interaction with the analytical system. Identity is a different question and may require complementary evidence such as mass spectrometry or another suitable technique. Researchers should record which question each method addresses rather than treating one result as a complete characterisation.
- Chromatographic purity: the relative distribution of detected peaks under the stated conditions.
- Identity: evidence that the material corresponds to the intended molecule.
- Quantity or content: evidence about how much target material is present.
- Biological effect: an experimental outcome that requires an appropriate model and controls.
Questions to ask about an HPLC-tested peptide
Is the report batch-specific? Is the chromatogram readable? Are the axes, principal peak and integration visible? Does the method provide enough context to understand the stated percentage? Is complementary identity evidence available? Does the receiving laboratory need independent incoming-material testing for the intended study?
